The captain's son, the future Dr. Brenner, attempted to recreate this phenomenon in the Nevada desert. But a disloyal lab worker pilfered lab samples to hand over to the Soviets, who were keen to start their parallel research. This spy in the mine shaft is the one who stole the materials.

The Briefcase's Contents

Given what is revealed about Brenner's work, the container most likely held some of the altered blood. In the scene, Henry is bleeding from his hand. He likely becomes contaminated upon opening the case, leading to the awakening of his powers—traits he would genetically impart to the kids Brenner would experiment on.

A second theory, the container could have held dark matter that eventually found its way to the Hawkins lab, where it maintains the stability of the mirror dimension.
